KIF18B通过调控细胞周期促进了三阴性乳腺癌的进展(英文)

摘要:

Triple-negative breast cancer (TNBC) is a highly aggressive cancer and a leading cause of cancerrelated mortality in women. Kinesin family member 18B (KIF18B) is a microtubule-based motor protein involved in a variety of cellular processes, such as cell division. KIF18B has been shown to affect the progression of multiple types of cancer. However, the association between KIF18B and TNBC is unclear. Through cancer genome atlas(CGA) database analysis, the result revealed that KIF18B was highly expressed in the TNBC tissues and the high expression indicated the poor prognosis of patients. Furthermore, the cancer cell line encyclopedia (CCLE) database demonstrated that KIF18B was overexpressed in TNBC cell lines. Then, KIF18B was knocked down in human MDA-MB-231 and HCC-1937 TNBC cell lines via transfection with a targeting short hairpin RNA. The results revealed that KIF18B knockdown inhibited cell proliferation, as demonstrated by the findings of colony formation and Cell Counting kit-8 assays. In addition, KIF18B knockdown led to cell cycle arrest at the G2/M phase through its involvement in the cell cycle pathway. The xenograft model further demonstrated that KIF18B knockdown inhibited tumor growth in vivo. These data indicated that KIF18B induced TNBC proliferation by regulating the cell cycle. KIF18B may be a potentially effective target for TNBC.
